http://www.wireshark.org/ -> Get Help -> Documentation -> [Scroll down to "Developer Documentation" and choose your preferred format.]
Also, don't forget about about the various README's in the wireshark top-level directory and within the doc/ subdirectory, in particular, doc/README.developer. Like all the Wireshark sources, it is viewable online from: http://www.wireshark.org/ -> Develop -> Browse the Code -> [Choose a branch, e.g., Trunk/] -> doc -> README.developer.
