Guy,
After much research I discovered that Microsoft have configured ISA Server 2004
to disable MTU Discover and limit the maximum MTU size to 576 (plus a header
would probably be 590).

Therefore my original concerns about a possible bug with Wireshark were
misguided and Wireshark is working as designed.
